Description

The CRH-binding protein inactivates corticotropin-releasing hormone (CRH; 122560), a potent stimulator of synthesis and secretion of preopiomelanocortin (176830)-derived peptides, thereby preventing inappropriate pituitary-adrenal stimulation in pregnancy (Potter et al., 1991).


Cloning and Expression

Potter et al. (1991) isolated and partially purified the binding protein and cloned and characterized its cDNA from human liver and rat brain. Expression of the cDNAs in COS-7 cells showed that the protein binds CRH with the same affinity as the native protein. Furthermore, the recombinant binding protein inhibited CRH binding to a CRH antibody and inhibited CRH-induced release of ACTH by pituitary cells in vitro.


Gene Function

Asakura et al. (1997) found that the thecal compartment of the human ovary contains a CRF system endowed with CRF, CRF1 (122561), and CRFBP.


Gene Structure

Behan et al. (1993) demonstrated that the human CRHBP gene consists of 7 exons and 6 introns.


Mapping

Behan et al. (1993) mapped the human CRHBP gene to chromosome 5q by Southern blot analysis of DNA from somatic cell hybrids. They mapped the murine counterpart to the distal region of chromosome 13 by means of analysis of interspecific backcross matings.

Studying somatic cell hybrids that contained various deletions of chromosome 5, Vamvakopoulos et al. (1995) regionalized the assignment of the CRHBP gene to 5q11.2-q13.3.
