Cloning and Expression

Using microarray analysis to identify transcripts upregulated in human umbilical vein endothelial cells (HUVECs) by TNF-alpha (191160), followed by database analysis and PCR of a TNF-alpha-treated HUVEC cDNA library, Mataki et al. (2000) cloned ZNF71, which they called EZFIT. The deduced 490-amino acid protein has 13 C2H2 zinc finger motifs. PCR or DNA chip analysis detected EZFIT in placenta and in adult and fetal brain, testis, pancreas, heart, small intestine, muscle, uterus, prostate, adrenal gland, peripheral blood leukocytes, macrophages, and monocytes, but not in liver, lung, colon, stomach, or in salivary or thyroid gland. Expression was also detected in stomach cancer and neurofibromatosis cells, but not in HUVECs without TNF-alpha treatment or in liver cancer tissue.


Mapping

Using database analysis, Mataki et al. (2000) mapped the ZNF71 gene to chromosome 19q13.4. Aubry et al. (1992) had erroneously mapped the ZNF71 gene to chromosome 22.
