Proline-arginine-rich end leucine-rich repeat protein (PRELP) is a connective tissue glycoprotein of the leucine-rich repeat (LRR) family (Bengtsson et al., 1995).


Cloning and Expression

Bengtsson et al. (1995) isolated the 58-kD PRELP protein from bovine articular cartilage and cloned the human PRELP cDNA from an articular chondrocyte cDNA library. Sequencing revealed that the gene encodes a 382-amino acid polypeptide with a calculated molecular mass of 42 kD. The protein sequence contains a putative secretory signal sequence, 10 leucine-rich repeats, several glycosylation sites, and a proline- and arginine-rich N terminus. It shares 36% and 33% sequence similarity with fibromodulin (600245) and bovine lumican (see 600616), respectively.

Grover et al. (1996) determined that PRELP is encoded by messages of 1.7, 4.6, and 6.7 kb, in descending order of expression level. The messages are abundant in juvenile and adult cartilage, but not present in neonatal cartilage nor in cultured fibroblasts. Among other adult tissues, PRELP is expressed in lung.


Gene Structure

Grover et al. (1996) determined that the PRELP gene spans approximately 16 kb of genomic DNA and contains 3 exons.


Mapping

Grover et al. (1996) mapped the PRELP gene to chromosome 1q32.
