PPFIA3, or liprin-alpha-3, belongs to the liprin-alpha gene family (Serra-Pages et al., 1998). See liprin-alpha-1 (LIP1, or PPFIA1; 611054) for background on liprins.
Serra-Pages et al. (1998) identified a family of genes encoding LIP1-related proteins, or liprins. The sequence of the predicted partial 474-amino acid liprin-alpha-3 polypeptide is 72% identical to that of LIP1. Serra-Pages et al. (1998) proposed that liprins are multivalent proteins that form complex structures that act as scaffolds for the recruitment and anchoring of LAR family PTPases. Northern blot analysis revealed that the liprin-alpha-3 gene is expressed as a 5-kb mRNA only in brain.
By screening human brain cDNAs for those encoding proteins larger than 50 kD, Ishikawa et al. (1998) identified KIAA0654, a liprin-alpha-3 cDNA. Since the first ATG was not within a Kozak consensus region, the KIAA0654 cDNA may lack some 5-prime sequences. The predicted 1,093-amino acid polypeptide shares 52% sequence identity with LIP1. The in vitro transcribed/translated product of the cDNA had a molecular mass of greater than 100 kD by SDS-PAGE. RT-PCR revealed that liprin-alpha-3 is expressed in several tissues.
By analysis of a radiation hybrid panel, Ishikawa et al. (1998) mapped the liprin-alpha-3 gene to chromosome 19.
Gross (2022) mapped the PPFIA3 gene to chromosome 19p13.3 based on an alignment of the PPFIA3 sequence (GenBank BC101518) with the genomic sequence (GRCh38).
