The HIRA (600237) protein shares sequence similarity with the S. cerevisiae Hir1p and Hir2p corepressors, which appear to act together on chromatin structure to control gene transcription. Lorain et al. (1998) stated that HIRA, like Hir1p, contains 7 N-terminal WD repeats and probably functions as part of a multiprotein complex. Using a yeast 2-hybrid screen with HIRA as bait, these authors identified HeLa cell cDNAs encoding HIRA-interacting proteins, or HIRIPs. The predicted partial protein sequence of HIRIP3 contained 20% acidic and 21% basic amino acids, including several stretches of lysine and arginine residues.
Lorain et al. (1998) found in in vitro studies that HIRIP3 interacted with HIRA and with H2B and H3 core histones (see 142711), suggesting to Lorain et al. (1998) that a HIRA-HIRIP3-containing complex could function in some aspects of chromatin and histone metabolism.
Gross (2012) mapped the HIRIP3 gene to chromosome 16p11.2 based on an alignment of the HIRIP3 sequence (GenBank AJ223349) with the genomic sequence (GRCh37).
