TEXT

Zinc finger proteins have been shown to interact with nucleic acids and to have diverse functions. The zinc finger domain is a conserved amino acid sequence motif containing 2 specifically positioned cysteines and 2 histidines that are involved in coordinating zinc. Kruppel-related proteins form 1 family of zinc finger proteins. See ZFP93 (604749) for additional information on zinc finger proteins.


Cloning and Expression

By screening a human monoblast cell line (U-937) cDNA library with a degenerate oligonucleotide based on a conserved region of Kruppel-related zinc finger proteins that encompasses the H/C link, Abrink et al. (1995) isolated cDNAs encoding 42 distinct Kruppel-related zinc finger proteins, including ZNF267, which they called HZF2. The partial ZNF267 cDNA contains almost an entire coding sequence, being only minimally incomplete at the 5-prime end. The predicted ZNF267 protein, which is composed of more than 733 amino acids, contains a nearly complete N-terminal KRAB domain, with both A and B boxes; a spacer region that is distantly related to the spacer regions of ZNF91 (603971) subfamily members; and a clustered zinc finger domain consisting of 1 zinc finger motif, followed by 3 degenerate fingers in which the zinc-binding ability has probably been lost, followed by 13 canonical zinc finger motifs. Northern blot analysis detected a major 4.0-kb ZNF267 transcript in HeLa cells, U-937 cells, and PMA-induced U-937 cells.


Mapping

The International Radiation Hybrid Mapping Consortium mapped the ZNF267 gene to chromosome 16 (RH104437).
