Using RNA fingerprinting, Dragoni et al. (1998) isolated a novel human endothelial cell cDNA designated EDF1 (endothelial differentiation-related factor-1). The EDF1 cDNA encodes a basic intracellular protein of 148 amino acids that shares 66% and 48% amino acid identity with the silkworm transcriptional coactivator MBF1 and the developmentally regulated Dictyostelium H7. Northern blot analysis detected expression of a 1.0-kb EDF1 transcript at varying levels in all adult and fetal tissues examined, with most abundant expression in adult heart, liver, and pancreas, and fetal kidney. When endothelial cells were induced to differentiate, there was a marked reduction of EDF1 as detected by Northern and Western blot analysis. EDF1 antisense RNA inhibited EDF1 expression and promoted endothelial cell differentiation. Dragoni et al. (1998) postulated that EDF1 may function as a bridging molecule that interconnects regulatory proteins and the basal transcriptional machinery, thus modulating the transcription of genes involved in endothelial differentiation, which is a crucial step in angiogenesis.
