Using the casein kinase II (CK2) alpha subunit (CSNK2A1; 115440) as bait in a yeast 2-hybrid screen, Bosc et al. (2000) cloned CKIP1 from a B-cell cDNA library. The deduced 409-amino acid protein has an N-terminal pleckstrin homology domain and a putative C-terminal JUN (165160) leucine zipper interactive domain. Northern blot analysis detected a major 1.5-kb transcript and a minor 4.4-kb transcript expressed at highest levels in skeletal muscle and heart. Intermediate expression was detected in brain, placenta, and lung, and weak expression was detected in liver, kidney, and pancreas. Fluorescence-tagged CKIP1 localized at the plasma membrane of transfected COS-7 cells. The plasma membrane localization was dependent on the presence of the N-terminal pleckstrin homology domain. In human osteosarcoma cells, CKIP1 localized within the nucleus and at the plasma membrane.
By coimmunoprecipitation and in vitro binding assays, Bosc et al. (2000) showed that CKIP1 interacted with full-length CSNK2A1 or with the N terminus of CSNK2A1. CKIP1 did not interact with CSNK2A2 (115442) or CSNK2B (115441). The immunoprecipitated proteins showed CK2 activity.
The International Radiation Hybrid Mapping Consortium mapped the CKIP1 gene to chromosome 1 (D1S3340).
