By sequencing clones obtained from a size-fractionated brain cDNA library, Nagase et al. (1998) cloned PCF11, which they designated KIAA0824. The deduced protein contains 1,644 amino acids. RT-PCR ELISA detected intermediate expression in all adult and fetal tissues and all specific brain regions examined.
De Vries et al. (2000) purified pre-mRNA cleavage factor IIm (CFIIm) of the 3-prime end processing complex (see 604978) from HeLa cell nuclear extracts and determined that it contains PCF11 and CLP1 (608757).
Crystal Structure
Meinhart and Cramer (2004) described the structure of a ser2-phosphorylated C-terminal domain peptide of RNA polymerase II (see 180660) bound to the C-terminal domain-interacting domain of PCF11. The C-terminal domain motif of ser2-pro3-thr4-ser5 forms a beta turn that binds to a conserved groove in the C-terminal domain-interacting domain of PCF11. The ser2 phosphate group does not make direct contact with PCF11, but may be recognized indirectly because it stabilizes the beta turn with an additional hydrogen bond. Iteration of the peptide structure results in a compact beta spiral model of the C-terminal domain. Meinhart and Cramer (2004) suggested that during the mRNA transcription processing cycle, compact spiral regions in the C-terminal domain are unraveled and regenerated in a phosphorylation-dependent manner.
By radiation hybrid analysis, Nagase et al. (1998) mapped the PCF11 gene to chromosome 11.
