Using a PCR-based probe to screen a human fetal liver cDNA library, Vallenius and Makela (2002) identified a novel cDNA encoding a deduced 401-amino acid protein with a molecular mass of 44.6 kD. The protein, which the authors designated CLIK1 (CLP36 (605900)-interacting kinase), contains a serine-threonine kinase domain, with the ser-thr active site signature between amino acids 223 and 235, 4 potential nuclear localization signals, and a proline-rich region in the N terminus. The full-length sequence shared only 20 to 30% identity with known serine/threonine kinases. Northern blot analysis of human adult tissues demonstrated a 7.0-kb transcript with strong expression in testis and low expression in all other tissues tested.
By Western blot analysis, Vallenius and Makela (2002) determined that CLIK1 undergoes autophosphorylation in vitro. Yeast 2-hybrid analysis showed that CLIK1 associates with the PDZ-LIM protein CLP36 (605900) through the LIM domain. Transfection studies demonstrated that CLIK1 localizes predominantly to the nucleus; however, cotransfection of CLIK1 with CLP36 demonstrated redistribution of CLIK1 to the cytoplasm and into actin stress fibers, where it colocalizes with CLP36. The presence of CLIK1 disrupted the periodic staining pattern of CLP36 in these fibers. Vallenius and Makela (2002) suggested that CLP36 specifically targets CLIK1 to actin stress fibers and that CLIK1 may represent a regulator of the actomyosin cytoskeleton in nonmuscle cells.
By sequence analysis, Vallenius and Makela (2002) mapped the CLIK1 gene to chromosome 20p13 between markers D20S117 and D20S113.
